The 2015-16 Under-21 Premier League is the fourth campaign of the new top tier of the Professional Development League. The league remains split into two, with both Middlesbrough and Reading promoted to the top division after finishing first and second respectively in Division 2 in 2014-15. They replace Fulham and West Ham, who were relegated.

In Division 2, Swansea City replace Bolton Wanderers ahead of their imminent recategorisation as part of this season's EPPP audit; Swansea will play Category 1 football this season in anticipation of being approved formally for this status, while Bolton applied for Category 2 instead of Category 1 status for the second cycle of the EPPP. Promotion and relegation of two teams will apply once again this season.
